    Complexity and maintenance : a comparative study of object-oriented and structured methodologies : a thesis presented in partial fulfilment of the requirements for the degree of Master of Business Studies in Information Systems at Massey University

    Maintenance has been found to be one of the most expensive phases in the life of an information system. It has been suggested that the use of object-oriented methods instead of traditional structured methods may be one way of reducing the cost of maintenance required for an information system. This thesis is an attempt to determine whether the object-oriented approach does in fact undergo a relatively smaller increase in complexity when subjected to a change in specifications than a similar system that is developed using a "structured methodology", and is therefore easier to maintain. The methodologies used in this study were Yourdon's (1989) Modern Structured Methodology and Booch's (1994) Object-Oriented methodology. The analysis phase of both methodologies were applied to the same case study twice in order to evaluate the effects of a change in the system's specifications. Once the two models for each methodology were complete, various metrics were applied to the structured system and a separate set of metrics were applied to the object-oriented system. The results of the models and the metrics were then analysed and validated in order to determine which system suffered a smaller proportional increase in complexity as a result of the changes to the system. It was found that overall, the object-oriented system proved to undergo a smaller increase in complexity, and it was therefore easier to maintain as a result of the changes than the structured system

    Repeated sub-concussive impact (e.g. soccer ball heading), a significantly lighter form of mild traumatic brain injury, is increasingly suggested to cumulatively alter brain structure and compromise neurobehavioural function in the long-term. However, the underlying mechanisms whereby repeated long-term sub-concussion induces cerebral structural and neurobehavioural changes are currently unknown. Here, we utilised an established rat model to investigate the effects of repeated sub-concussion on size of lateral ventricles, cerebrovascular blood–brain barrier (BBB) integrity, neuroinflammation, oxidative stress, and biochemical distribution. Following repeated sub-concussion 3 days per week for 2 weeks, the rats showed significantly enlarged lateral ventricles compared with the rats receiving sham-only procedure. The sub-concussive rats also presented significant BBB dysfunction in the cerebral cortex and hippocampal formation, whilst neuromotor function assessed by beamwalk and rotarod tests were comparable to the sham rats. Immunofluorescent and spectroscopic microscopy analyses revealed no significant changes in neuroinflammation, oxidative stress, lipid distribution or protein aggregation, within the hippocampus and cortex.     A method for the identification of chromosomal inversions is described. Single-stranded sister chromatids are generated, for example by CO-FISH. A plurality of non-repetitive, labeled probes of relatively small size are hybridized to portions of only one of a pair of single-stranded sister chromatids. If no inversion exists, all of the probes will hybridize to a first chromatid. If an inversion has occurred, these marker probes will be detected on the sister chromatid at the same location as the inversion on the first chromatid

    Mid-Infrared High-Contrast Imaging of HD 114174 B : An Apparent Age Discrepancy in a "Sirius-Like" Binary System

    We present new observations of the faint "Sirius-like" companion discovered to orbit HD 114174. Previous attempts to image HD 114174 B at mid-infrared wavelengths using NIRC2 at Keck have resulted in a non-detection. Our new L'-band observations taken with the Large Binocular Telescope and LMIRCam recover the companion (ΔL\Delta L = 10.15 ±\pm 0.15 mag, ρ\rho = 0.675'' ±\pm 0.016'') with a high signal-to-noise ratio (10 σ\sigma). This measurement represents the deepest L' high-contrast imaging detection at sub-arcsecond separations to date, including extrasolar planets. We confirm that HD 114174 B has near-infrared colors consistent with the interpretation of a cool white dwarf (JLJ-L' = 0.76 ±\pm 0.19 mag, KLK-L' = 0.64 ±\pm 0.20). New model fits to the object's spectral energy distribution indicate a temperature TeffT_{\rm eff} = 4260 ±\pm 360 K, surface gravity log g = 7.94 ±\pm 0.03, a cooling age tc_{c} \approx 7.8 Gyr, and mass MM = 0.54 ±\pm 0.01 MM_{\odot}. We find that the cooling age given by theoretical atmospheric models do not agree with the age of HD 114174 A derived from both isochronological and gyrochronological analyses. We speculate on possible scenarios to explain the apparent age discrepancy between the primary and secondary. HD 114174 B is a nearby benchmark white dwarf that will ultimately enable a dynamical mass estimate through continued Doppler and astrometric monitoring. Efforts to characterize its physical properties in detail will test theoretical atmospheric models and improve our understanding of white dwarf evolution, cooling, and progenitor masses.Comment: 6 pages, 3 figures, to be published in the Astrophysical Journal Letter

    Repeated long-term sub-concussion impacts induce motor dysfunction in rats : a potential rodent model

    Whilst detrimental effects of repeated sub-concussive impacts on neurophysiological and behavioral function are increasingly reported, the underlying mechanisms are largely unknown. Here, we report that repeated sub-concussion with a light weight drop (25 g) in wild-type PVG rats for 2 weeks does not induce detectable neuromotor dysfunction assessed by beamwalk and rotarod tests. However, after 12 weeks of repeated sub-concussion, the rats exhibited moderate neuromotor dysfunction. This is the first study to demonstrate development of neuromotor dysfunction following multiple long-term sub-concussive impacts in rats.     Differentiated State of Initiating Tumor Cells Is Key to Distinctive Immune Responses Seen in H-Ras

    Heterogeneity in tumor immune responses is a poorly understood yet critical parameter for successful immunotherapy. In two doxycycline-inducible models where oncogenic H-RasG12V is targeted either to the epidermal basal/stem cell layer with a Keratin14-rtTA transgene (K14Ras), or committed progenitor/suprabasal cells with an Involucrin-tTA transgene (InvRas), we observed strikingly distinct tumor immune responses. On threshold doxycycline levels yielding similar Ras expression, tumor latency, and numbers, tumors from K14Ras mice had an immunosuppressed microenvironment, whereas InvRas tumors had a proinflammatory microenvironment. On a Rag1-/- background, InvRas mice developed fewer and smaller tumors that regressed over time, whereas K14Ras mice developed more tumors with shorter latency than Rag1+/+ controls. Adoptive transfer and depletion studies revealed that B-cell and CD4 T-cell cooperation was critical for tumor yield, lymphocyte polarization, and tumor immune phenotype in Rag1+/+ mice of both models. Coculture of tumor-conditioned B cells with CD4 T cells implicated direct contact for Th1 and regulatory T cell (Treg) polarization, and CD40-CD40L for Th1, Th2, and Treg generation, a response not observed from splenic B cells. Anti-CD40L caused regression of InvRas tumors but enhanced growth in K14Ras, whereas a CD40 agonist mAb had opposite effects in each tumor model. These data show that position of tumor-initiating cells within a stratified squamous epithelial tissue provokes distinct B- and CD4 T-cell interactions, which establish unique tumor microenvironments that regulate tumor development and response to immunotherap
